Targeted delivery of small interfering RNA: approaching effective cancer therapies.
Three of the primary requirements for the development of effective dual-targeting therapeutic modalities for the treatment of cancer are the tumor-targeted delivery of the therapeutic molecules of interest to the tumor site(s) in the body (both primary and metastatic), passage of the molecular therapeutic through the cell membrane, and targeting specifically a growth or apoptotic pathway. Howev...
متن کاملTargeted Delivery of FLT-Morpholino Using Cyclic RGD Peptide
PURPOSE We previously showed that intravitreal injection of the sFLT morpholino-oligomer (FLT-MO) suppresses laser-induced choroidal neovascularization (CNV) in mice by decreasing the membrane bound form of Flt-1 while increasing the soluble form of Flt-1 via alternative splicing shift. In this study, we examined whether cyclic RGD peptide (cRGD) can promote morpholino-oligomer accumulation in ...
متن کاملSmall interfering RNA; principles, applications and challenges--
Gene silencing using RNAi (RNA interference), has recently been used as a successful laboratory technique in determining the function and control of gene expression and provides a wide range of applications in molecular biology and gene therapy. RNAi is a method of suppressing gene expression. In this direction, a single-stranded RNA molecule of about 21–23 nucleotides, called siRNA (small inte...
متن کاملFunctional Nanostructures for Effective Delivery of Small Interfering RNA Therapeutics
Small interfering RNA (siRNA) has proved to be a powerful tool for target-specific gene silencing via RNA interference (RNAi). Its ability to control targeted gene expression gives new hope to gene therapy as a treatment for cancers and genetic diseases. However, siRNA shows poor pharmacological properties, such as low serum stability, off-targeting, and innate immune responses, which present a...
متن کاملConjugated polymer nanoparticles for small interfering RNA delivery.
Loosely aggregated conjugated polymer nanoparticles (CPNs) were used as nontoxic and efficient small interfering RNA (siRNA) delivery vehicles with delivery visualization. A significant down regulation (94%) of a target gene was achieved by transfection of HeLa cells with the CPNs/siRNA complexes, supporting CPN as a promising siRNA delivery carrier.
